I know that slope of line = (y2-y1)/(x2-x1). But i am confused, how we select the term y2 ,y1 x2 and x1 for solving, when i got R(4, 1.5 ) and Q(-2, -3). Is it necessary R(x2, y2 ) and Q(x1, y1).

not at all. no one says which one is "first" and which one is second. you just have to be consistant

I think it doesn't matter which point you assign x1,y1 or x2, y2 since we are mainly interested in the difference between the two points in question. You just need to be consistent.

In fact, (-3-1.5)/(-2-4) should be equal to (1.5+3)/(4+2)
