What is amino acid subsitution?

It could be related to postranslation modification to the side chain of the amino acid. Such as Proline could be modified to become hydroxyproline [a presence of an -OH and participate in hydrogen bonding thus making the protein more stable]. Acylation is adding an fatty acid chain usually where sulfur are found within the side chain such as cysteine. There are also phosphorylation, proteolysis, disulfide bonds, etc.
