they are basically made in the cytoplasm, a lot of organelles work on making them; but gene proteins play the major role.

Enzymes, like all proteins, are synthesized by ribosomes. The cell 'decides' to synthesize enzymes in response to some environmental stimulus: it needs more of the enzyme to catalyze some reaction and that condition favors transcription of genes which code for the specific enzyme. What controls the enzyme is a little sketchier. Most enzymes have multiple levels of control: the first, crudest one is the amount of a specific enzyme the cell requires and then makes. Finer control is offered by post translational modification - usually phosphorylation and dephosphorylation, but occasionally modification with less common groups - which affects the structure of each individual copy of the enzyme, and hence it's ability to bind its substrate or catalyze its reaction. Scientists - and teachers grading homework - usually refer to this as 'activation' or 'inactivation' of the enzyme. Beyond that, many enzymes are also under 'substrate level control' where the reaction they catalyze depends on the relative concentrations of substrate and product and so are very responsive to intracellular conditions.
Okay, blue's answer is a tad bit high class :P Easily put, the enzyme proteins in the cells are made by the ribosomes, the energy for it comes from the mitochondria, and the direction from the genes. How does the gene direct it to work, well, somebody here in openstudy made me understand a long time ago that the shape of the gene plays a role here, like a lock and key mechanism. So, when a specefic gene is making an enzyme, its shape ensures that you get the enzyme it is making and not a blob of protein. But yes, I could be wrong.
