In DNA replication what is a 3' & 5' end ?

The DNA backbone is asymmetric. The 3' end of the strand has an unbound hydroxyl (OH) group on the 3' carbon whereas the 5' end has an unbound phosphate group on the 5' carbon of the deoxyribose.
