Each amino acid is carried to the ribosome by a tRNA which has an anticodon which binds to the mRNA. The amino acids are specified by triplet codes in the mRNA known as codons. The anticodon binds to the codon, ensuring the correct amino acid is delivered. Most amino acids are specified by more than one codon, so a slight difference in sequence might not lead to a change in protein sequence because the amino acid specified might not change. Exceptions to this rule are methionine, which is only specified by ATG in the DNA or AUG in the mRNA. Methionine is the first amino acid in most proteins so the open reading frame (ORF) ususally starts ATG.
