What is a limitation of using electron microscopes to view specimens?

Electron microscopes' disadvantages are : . They can't operate unless in a complete vacuum . Local vibrations, radiations and magnetic fields have to be reduced at minimum . They need very stable and high voltages to work properly (even 5kV) . Futhermore, they are expensive as hell. BUT they grant an incredible definition and 3D-wiew...

Additionally the specimen must be killed (if we're talking about cells) and prepared on a slide. You can't view live cells with it.
