Well a stain is something that will bind to a substance based on size, charge etc in histology Eosin is an acidic dye and has the basic couterpart Haematoxylin therefore dying the charged nucleus blue and cytoplam red based on charge A reagent is a substance used in a chemical reaction to detect, measure, examine, or produce other substances. so we add a reagent to collegen etc and it will react with it and form a compound with it which will show it up like formaldehyde
