e = mc^2 basically says that the rest mass of any given mass can be transformed into some quantity of energy. A common analogy to describe this is thinking of the equal sign as a "tunnel": Mass can go through the "tunnel" and come out as energy. This is seen commonly in nuclear reactions such as nuclear fission and fusion, where an atom is split or two atoms are smashed together, and the mass of roughly 1/5th of a proton is "missing" afterwards. This mass was transformed into pure energy in the reaction.
