If you are solving an equation like 3(2x-1) =27+x do you not pay attention to the parentheses? Also how would you begin to solve it?

You use distribution to deal with the parentheses. 3(2x-1)=6x-3

Distribute your 3 and then get x by itself to solve.

Do I start with the order of operations to get x by itself?

Order of operations only pertains to "operations", while it is important, it does not help you isolate 'x'. Start as @nbouscal suggested and distribute the 3 into the quantity and when you rewrite the equation the next step should be more obvious.
