I wonder how something like TeX or MS Word could translate some marked-up file into all the fonts and formatting and stuff.

Generally, if your given word documents, they'd need to be laid out in signatures with a pagination program. Signatures are the layouts that will actually run on the press, and then (more often than not) be folded down to be assembled (collated) with other signatures, joined and three edge trimmed.
