The movie from 1953 is missing some scenes and parts from acts such as when Portia stabs herself in the thigh to prove to Brutus that she is strong enough to keep his secrets. Another scene that was changed is when Caesar was being killed. All the conspirators ended up cutting one another along with killing Caesar but in the movie only Caesar was stabbed.

For the 2012 movie:

The movie shows Caesar's entire life, but the book only shows the events leading up to his death. Cleopatra isn't even mentioned in the book, but is an essential character in the movie. Pompey is mentioned briefly in the first act of the book, but is given a major part in the movie.
