The test charge is just a small (usually) electric charge, like an electron. It's purpose is to 'feel' the electric field and react to it. Think of the case of gravity. A test charge for gravity would be some small mass, like an apple, that could react to the earth's much bigger gravitational field. This way we can test the effects of the Earth's gravity without compromising the situation. For example if we used the moon as our mass to check our gravitational equations we'd be in for trouble because the moon has its own field that we cannot ignore. Now the gravitational field of the Earth would be distorted by the moon's large field. We use something small because its own gravitational field is negligible. A test charge like an electron is not going to change the electric field significantly, and we can thus ignore its effects on the situation.
Does we always place the Test Charge\[^qo\] on that particular point where the original charge \[Q\] is, for testing Electric field strength?

No, in fact we do not place it at the original point, we place it where we want to test the field. For example, if we wanted to know the charge of Q we could put a test charge one meter away from Q, the because we know the value of the test charge q0, we could see the force acting on q0 and calculate Q with the equation \[F= 1/(4pi \epsilon _{0})(Q*q _{0})/R ^{2}\] Where R is the distance (one meter in this example). This test would let us find Q indirectly. Since electrically charged particles are the only things that respond to electric fields, test charges are required to see the effect of the field.

Test charge is just imaginary. So the "note" at the end of your text is irrelevant. Besides, if an actual BIG charge was used, the result would not be modified, so this note is really nonsense.
