Depends on what you mean by block. If you click on someone's avatar or username, you should get a tooltip after a second that has a block user button. You can also access it on their profile. Blocking a user this way will make it so:
 - You are not notified when they mention you in questions or replies.
 - They cannot send you a private message.
 - They cannot fan you (and thus give you a fan testimonial).
 - You cannot fan them (and thus give them a fan testimonial).

If someone is doing something that's bothering you, you also can and should report them abusive. There are report abuse links on every question and reply, as well as every private message and chat (you have to hover over a chat to see the report abuse link).

If that's not what you had in mind, you probably can't do it, but I'm still curious to know what you want blocking to do if the above doesn't line up with those expectations.
