Are there any cell-made proteins that do not need ER vesicles to be transported to their designated location? If so, what kind are they?

Yes . 1.cytoplasmic, 2.nuclear , 3.organellar (mitochondria and chloroplast) 4 peroxisomal proteins dont need ER vesicles for their transportation. Proteins translocated to these organelles have a signal sequence either at their N or C termini that target them to the respective organelle .
