What is a limitation of using electron microscopes to view specimens?

Your samples have to be dead and therefore no live samples have every been viewed, no time lapse can occur and all you can see is structure, not while it is moving about. It is also rather expensive and the process to set up a specimen for an EM, if messed up, will ruin your sample. Transmission only lets you see a "slice" of your specimen and if you don't know what you are looking for (or at) it is useless.
