Hi everyone, I need your help for some calculations. I need to work out the recipe for making 50ml of 2x sample buffer (Composition of 2x = 20% glycerol, 100mM TrisCl pH 6.8, 4mM EDTA pH 8, 4% (w/v) SDS, 200mM DTT, 0.001% (w/v) bromophenol blue) given that I have been provided with the following stock solutions and salts: 100% glycerol 1M Tris pH 6.8 0.5M EDTA pH 8.0 SDS powder DTT powder (mw=154) 0.25% (w/v) bromophenol blue sterile water. Can you please explain to me your workings. Thanks.

I think that I've got it. Glycerol = 10ml of 100% glycerol 1 M Tris pH 6.8 = 5ml 0.5 EDTA pH 8= 0.4ml SDS powder = 2g DDT powder = 1.54g bromophenol = 0.2ml and water = 34.4ml
