Astrophysics and planetary physics.  You can only determine the composition of the atmosphere of Jupiter, or how much oxygen has been manufactured by distant stars, through spectroscopy.

Organic chemistry, including medicinal chemistry.  It's extremely difficult to know exactly what molecule you just made without the subtle tools of spectroscopy, which can tell you exactly where all the atoms are.
