It's pretty simple. Do you know about RNA polymerase? It's an enzyme. A protein, whose job is to read DNA strands, and make RNA out of them. And that's called "transcription." Here's a picture of transcription in action: http://www.glogster.com/media/4/29/17/50/29175041.jpg You see that big, almost clear bubble? That's the RNA polymerase. And you see that dark blue strand of DNA, on the bottom row? That's the DNA strand that the RNA polymerase is reading. The RNA polymerase sweeps across a strand of DNA, and as it sweeps along, it makes RNA. You see that red strand? That's the RNA that's being made, by the RNA polymerase. The RNA polymerase sweeps across a strand of DNA, and it reads the letters. You know how base pairs work, with DNA to RNA, right? T goes to A A goes to U G goes to C C goes to G For example (you can see in the picture), everytime the RNA polymerase sees the letter A, it will make the letter U. Everytime it sees the letter T, it will make the letter A. And it does this for the entire DNA strand. Do you get it?

Sigma factor is only needed for RNA initiation. And I think that only prokaryotes use it. Like bacteria.

Yes. That's called "translation."
i don't thinks translation is replication... replication is process of synthesis of new strands... so can we consider translation and transcription in this replication process?
Oh I'm sorry. Translation is how proteins are manufactured. You meant to ask how proteins are replicated. I read that wrong. I'm not sure exactly how proteins replicate. I don't even know if normal proteins can replicate. I have heard that prions (harmful proteins) can replicate.
