Remember that amino acids are coded by codons ie groups of three nucleotides. If the correct sequence is coded then the open reading frame is said to be "in frame". The open reading frame is the sequence of codons without a stop or terminator codon. One nucleotide more or less can mean that the wrong amino acids are coded - the reading frame is out of frame, or shifted. I hope that's enough of a clue! best wishes
