i have a question regarding Sanger sequencing of DNA..We add primer initially to provide nucleotides chain for DNA polymerase to start the synthesis.But how are we going to know what primer of what nucleotides sequence is going to fit in without knowing DNA sequence itself.

We have a RNA primer in Transcription.There is nothing like we have different types of primer for different nucleotides.
