I would like to emphasize a word. May I do that by hyphenating it? For example, the word, P-L-E-A-S-E.

It depends on what you are writing. Online, hyphenation would work. In a formal paper, this would probably be unacceptable. A word can be emphasized with italics or with repetition or exclamation marks. My teachers always tried to help me make emphasis clear through the way I wrote.
