I believe that the eyes of Dr. Eckleburg's demonstrate that nothing goes unnoticed. The location of the sign in the Valley of Ashes (or whatever it is called) is valuable because when Myrtle was killed by Daisy, the truth was revealed to Tom yet Gatsby took the blame for it due to his hatred for Gatsby. The theme of the decline of the American Dream relates to the eyes of the doctor because the watchfulness can easily signify that some people do notice things that otherwise would go unnoticed such as a death that is representative of the dream.

Despite the lack of concrete evidence in the book, the eyes of Dr. Eckleburg have generally come to represent the eyes of god, staring down and judging the decline of American morality and the increased materialism of the time.

This is symbolized most strongly when Wilson sees the eyes as those of gods, driving him insane and pushing him to kill Gatsby.
