Okay lets start with DNA. DNA is the material that make you a human for example. In order to investigate the relation between organisms we can compare the DNA of the organisms and see if there are any relationship the organism if the DNA is very much alike. For example the difference in humans and bonobos genes, is determined by only 2 %. The remaining 98 are identical. For proteins we can use a similar method. You can might remember that DNA is being translated into proteins (after a longer process). Now lets for example think about hemoglobins if we set humans as a reference we can observe if there are changes in the amino acid sequence or a base have been change ect. This way can determine if we are closely related.
