Explain why a plasmid and a segment of DNA to be inserted into a plasmid must be cut with the same restriction enzyme

OpenStudy (shrutipande9):

restriction enzymes cut at a specific site. ill give u an eg. a restriction enzyme ECOR1 cuts the dna fragment after G at site 5'GAATTC3' 3'CTTAAG5' so the cuts obtained would be5'G AATTC3' 3'CTTAA G5' This is a staggered cut. if the same enzyme is used to cut the segment of DNA to be inserted then the overhangs created would be same ie ttaa. so it would be easier to ligate the segment to d plasmid.
