Why do macromolecules differ in the amount of energy they contain?

this could be answered far more in detail in a more advance course, such as biochemistry, but basically, different molecules have different bonds, which carry more or less energy. They vary by carbohydrates, fats and lipids, which all carry varying amounts of energy.
