Explain why a plasmid and a segment of DNA to be inserted into a plasmid must be cut with the same restriction enzyme.

most of the restriction enzymes give stagerred cuts giving overhangs...so for the plasmid and segment of DNA to be ligated...the overhangs created must base pair. so if both are cut with the same restriction enzyme then the over hangs produced would be complementary and can base pair very well.
