He cut away the pollen bearing male parts of a flower and dusted that flower from pollen from another plant.

By doing this, he was able to selectively cross-pollinate pure bred plants with particular traits and observe the outcome over many generations. This was the basis for his conclusions about the nature of genetic inheritance.
