Eukaryotic cells are much larger and much more complex than prokaryotic cells. Eukaryotic cells have a true nucleus, while prokaryotes do not. Eukaryotic DNA is linear, while prokaryotic DNA is circular. Eukaryotic cells are filled with a large, complex collection of organelles, many of them enclosed in their own membranes, while prokaryotic cells contains no membrane-bound organelles that are independent of the plasma membrane. This is a very significant difference, and the source of the vast majority of the greater complexity of the eukaryotic cell.

significant differences between prokaryotes and eukaryotes at the molecular level. 1. the genome of prokaryotes comprises a single covalently closed circular DNA molecule/chromosome whereas in eukaryotes the genome is fragmented into a number of chromosomes. 2. the DNA of prokaryotes is complexed with non-histone proteins and lacks higher order packaging whereas in eukaryotes the DNA is complexed with histone proteins organized into higher order structures like nucleosome , solenoid rod etc 3.the ribosomes of prokaryotes are of 70S type whereas those of eukaryotes are of 80S ( but organellar ribosomes of eukaryotes are of 70S type) 4. the single chromsome in prokaryotes has only one origin of replication whereas in eukaryotes every single chromosome contains multiple origins of replication. 5.prokaryotes have a single RNA polymerase whereas eukaryotes possess 3 different types of RNA polymerases. 6.prokaryotic genes lack introns whereas eukaryotic genes possess introns. 7. Genes in prokaryotes are generally organized into operons which is uncommon in eukaryotes. 8.the start codon in prokaryotes codes for formyl-methionine(fMet) whereas in eukaryotes it codes for methionine. 9.proteins synthesized by prokaryotes lack post-translational modification like phosphorylation , glycosylation etc which are present in eukaryotic proteins.
