I could not find anything about the Tuskegee Institution being Black Separatists, but the UNIA was Black Separatist as well as Black Nationalist. Yes, and yes the Tuskegee Institution is an influential University of Blacks, at least it used to be. It was built in 1881 aka Reconstruction Era.
