they looked for adenine, cytosine, guanine, and thymine. thymine only occurs in dna in rna there is uracil instead. They also discovered rna and mrna

The main objective of the project was to know the complete sequence of human genome. The gene identification is a complicated thing which involve identification of exon, promoters, reading frames etc. most of the human-gene's positions were already identified even before the complete sequencecing: using classical genetics and modern molecular biological techniques
