Why does the enzyme pepsin (present in the stomach) denature in the intestine? The enzyme denatures due to change in temperature as it enters the intestine. The enzyme denatures due to the alkaline environment of the intestine. The enzyme denatures due to lack of substrate molecules in the intestine.

Once out of the stomach the Ph of the contents is raised by the Bile to a copable Ph level for the intestinal region. As Pepsin Denatures at pH5 (or more like pH6.8) pepsin works in acidic medium ( its optimum pH being 2) it is denatured at pH above 4 and the pH of intestinal juice is alkaline so it stops working as the chyme enters duodenum as acid( HCl) is nuetraliz or your second choice
