Down's Syndrome is properly called "Trisomy 21." Ordinarily each human cell has two copies of each chromosome - one from each parent. In trisomy 21, as the name suggests, they get an extra (i.e., third) copy of the 21st chromosome. Which causes Down's Syndrome. That is the defining, causative feature of the condition (note that I don't call it a disease). So there is really only one way for it to happen.
