Calculation: Transformation efficiency - gene cloning Competent Escherichia coli cells were transformed with 5 µlof 5 ng/µl of closed circular pGEM plasmid DNA in a total volume of 1 ml of culture medium. The plasmid pGEMconfers resistance to ampicillin. 200 µl of the 1 ml mixture was plated onto an ampicillin-containing agar plate to select for transformants carrying the pGEM plasmid. Upon overnight incubation of the plate, 200 colonies were counted. Calculate the efficiency of this transformation experiment, expressed as number of colonies per microgram (µg) of plasmid DNA.
