Restriction enzymes are used to cleave both the insert and vector DNA at specifc restriction sites. The cleaved insert and vector molecules are mixed and allowed to anneal, forming a hybridized construct. DNA ligase is then added to catalyze the formation of the phosphodiester bonds between the DNA molecules to form the hybrid complex.
