when mRNA is being translated in the ribosomal complex. tRNA's carrying monomer peptides enter the large subunit of the ribosome. as the ribosome 'reads' the mRNA message each peptide is added to a growing chain held by sulfo-diester covalent bonds. The completed protein begins naturally folding and forming its secondary structure. It then must be transported to the golgi and (sometimes) added to a chaperonin to create the right chemical conditions for tertiary structure to properly form. Additional proteins can be added to this protein after it is fully formed, thereby causing quaternary structure.
