I need help designing a durable, eco-friendly, cost efficient, waterproof bag that has a stable closing mechanism. This bag will be used to cover up about 60% of a pothole. This bag has to contain factors of biomimicry. Any suggestions?

you could try a large zip lock bag.  That should give you what you need and if you live near an Ikea, they have large eco-friendly bio degradable zipper bags.

The closing mechanism wouldn't be very efficient because if a car runs over it, either the internal liquid will burst out or ooze. Also, the plastic has to last for eat least 6 months.
