OpenStudy (frostbite):

Could somebody help me making a figure a bit more nicely than my handsketch? @ParthKohli

OpenStudy (frostbite):

Handsketch attached.

OpenStudy (frostbite):

I know my handwriting is epic and not understandable at all :)

OpenStudy (frostbite):

@Somy

OpenStudy (somy):

owh my

thomaster (thomaster):

you mean making it digitally? or improve your drawing skills :P

OpenStudy (frostbite):

Digitally :P

OpenStudy (frostbite):

What do you mean??! I got epic drawing skill§!

OpenStudy (frostbite):

basically what happens is that: an R-loop (good ilustration on google), is being cut by a protein called Cas3, inside a huge protein complex called "Cascade". Next Cascade dissociate, and another single stand is being cut by Cas3. Last step is Cas3 is cutting small DNAs while unwinding double stand DNA. That is the basic principle, and it is a bit complicated yes, biology knowledge is a huge advantage :)

OpenStudy (somy):

lol @thomaster sadly i can't do that @Frostbite

OpenStudy (frostbite):

I've been trying my self in illustrator.. the results were catastrophically.

thomaster (thomaster):

ehh, drawing software? Even in paint it's easy to draw this. If you want a professional figure you'd need something better, photoshop-like software.

OpenStudy (frostbite):

@thomaster yeah I tried as said adobe illustrator. Do you think you can remake it AND improve the drawing a bit? :)

OpenStudy (somy):

good luck @thomaster lmao i would do it if i could

thomaster (thomaster):

you want it to look like this? http://www.mdpi.com/ijms/ijms-14-14518/article_deploy/html/images/ijms-14-14518f1-1024.png (this is probably not the same as what happens in your description but I mean the way they drew things here)

OpenStudy (somy):

owww this is good

OpenStudy (frostbite):

Yeah kinda, just in a little other design way?

OpenStudy (frostbite):

Or in fact exactly like that, just with my steps.

OpenStudy (somy):

edit them then

OpenStudy (somy):

i don't quite understand your drawing

OpenStudy (frostbite):

Is it the last step that is causing trouble? What happens is that Cas3 is doing exonucleolytic cutting of the DNA in the 3' -> 5' direction while unwinding the duplex. The 5' -> 3' stand is being handled by endonucleolytic cuts, and then 3' -> 5' exonucleolytic cutting of the new stand.

OpenStudy (somy):

hmmm i'll try doing them, but it'll take a loooooooooooooot of time

OpenStudy (somy):

when do you need it to be done?

OpenStudy (frostbite):

Wednesday the latest :)

OpenStudy (somy):

lol okay i'll try doing it, by i don't promise that it'll look amazing or so

OpenStudy (frostbite):

Any help is very appreciated, I could really use it.

thomaster (thomaster):

I made this in paint :P

thomaster (thomaster):

but with some more time I could do way better

OpenStudy (somy):

good jooob!! @thomaster lol now he can do it in no time

OpenStudy (frostbite):

@thomaster Darn paint is better than me! :( But it looks awesome! Think you could make you version of the full figure?

thomaster (thomaster):

haha I just made that for fun. I have to work now but I'll give it a show when I'm back.

OpenStudy (frostbite):

That would be great, thanks.

OpenStudy (somy):

OpenStudy (somy):

whats written in the circle that i made?

OpenStudy (frostbite):

In the picture ef.jpeg it is all the same protein Cas3 working on the DNA. in the 539ee....jpeg it say "crRNA"

OpenStudy (frostbite):

The triangle shapes I made to refure to helicase activiy (unwinding of DNA duplex), and the circle ones are nuclease activity.

OpenStudy (somy):

here you go

OpenStudy (frostbite):

Look great! :) Thank you so much

OpenStudy (somy):

you are welcome :) @Frostbite
