Hopkins thought that there must be something in normal food which was not represented in a synthetic diet made up of pure protein, pure carbohydrate, fats and salts; but the nature of that something was unknown to him.” To test this he fed Young rats on such synthetic diets and found that they failed to grow and even lost weight unless they were given small amounts of milk (a natural diet) daily. Hopkins concluded that milk contains “accessory food factors,” which are required only in trace amounts but are indispensable for normal growth and maintenance. This finally led to the formulation of "Vitamin hypothesis".
Now since he wanted to test effects on growth, it's more reasonable to use young rats rather than adult ones because growth in adult rats is not very much significant !
