Substrate acts as the missing puzzle piece for an enzyme. An enzyme needs to bind to its corresponding substrate.

Therefore: the more substrate that is available, the higher the velocity. HOWEVER, at a certain point there will be no more enzymes able to fill any excess substrate. Hence, the velocity will plateau at a maximum and look something like the drawing.
