Hello. May I ask have you ever done an Enzyme Lab before? If yes please click on the link below to help me. Thank you!
Yes I have worked in a lab with enzymes, mostly regarding biophysical characterization and kinetics.
Thank yo for your soon reply. But have you done the lab that I need help with?
@Frostbite
Of cause I do not do your work, I may guide you and help you in specific areas in your problem. But I won't make your report.
Yes. I definitely understand that. But do you know the answers of it? And are you sure your answers were correct?
I am sure yes.
And I can also argue for my statements obviously.
I should perhaps say I am educated as a biochemist and protein scientist.
Oh wow...I see. So, can we start yet? Now, I will submit my answers to you to see if I am correct. You just need to correct me? Is is OK? Thanks
Sure.
and I look into it.
Yes. That's more convenient. Thanks. I will choose to send to email. Thanks
Please wait for me 1 min.
Yes. I am just sent it to you. Please check it. Many thanks
In general it all looks correct. But I do got some comments. First of all you write the following for a hypothesis "If the higher concentration of the enzyme will increase the rate of the reaction then the absorbance also will increase" You talk about absorbance (quantitative measurement), but I do not see those results anywhere. So think fast about, what kind of experiments you are pulling of and what kind of data they produce. Are the data you produce quantitative or qualitative?
It is trying to answer what the antioxidant does for the human body.
No it got something to do with your hypothesis, to talk about absorbance we need a number if used quantitative. What I am saying is that you are doing a qualitative experiment: You get out some results to which the answer is: Yes or No. So your hypothesis isn't to check the absorbance as an indicator of reaction rate.
Sorry...I'm stucked....Can you think of any correct hypothesis?
Sure I can :) not to worry, I send back a revised version. But I just also write here.
I'm very bad at doing a lab, so I always need an expert like you to help me with.
That's so wonderful. my expert. You have save me! ...
And if I make any mistakes, please fix it to me. I'm always willing to study from you and my mistakes too.
Btw: am I under time pressure or can I write relaxed?
You can write with relaxed time, expert. Just to make sure the answers is correct. That's the best. Take your time
The enzyme observation report thingy, I can't open unfortunately, so I gotta trust what you do there is correct, or I can simply explain what I would (with very simple materials that is present in every house hold)
sorry... I will resend it to you? Is it OK?
You can check your mail now
Can you open it?
I'm not sure. It looks like I'm look at the same document as the assignment given. How about I just look at that last... otherwise I might get confused?
oh Ok...Thanks
Hello, Oops..I am misunderstood. Yes, I already sent to you the Enzyme Observations Form. Please check it
@KittyT I am back now, and got time to look upon it again. Hope the first corrections was something to work with.
Hello @Frostbite Are you there?
Yes I am here.
@FrostBite Hello. I will redo my work and then I will email you again. So, we can check it together. OK? Thank you for everything
@KittyT Yes I'll be happy to look at the new version ones you've redone it. I hope the corrections are somehow useful. I tried to hold it as scientific as I could, but also as easy as possible to fit your level. :)
@KittyT right here I mean yes.
Ok..Let's start
Okay lets start with the hypothesis. You wrote about absorbance, but I got to ask: have you at any point in this experiment worked with absorbance?
I don't know. Can you open the instructions of this lab again? I'm suck at this
Well let me ask it like this then: did you use a special kind of machinery for this kind of experiment (cause you need a special machine to obtain absorbance results)?
No it doesn't look like it form the instructions part. Okay so to remake a hypothesis. Our hypothesis is what we expect from the experiment / what do we wish to test. And I think it would be good say something like: "Can liver break down hydrogen peroxide and what products are being formed?" How does that sound to you?
"Can liver break down hydrogen peroxide and what products are being formed?" This will be the hypothesis.. Can a hypothesis be a question? Or it have to start with If...then?
Where I work we don't mind writing it to as a question (cause the hypothesis is OUR thoughts and are to guide us in our research). Although we can write it as a statement. But we need some more info then. Do you remember the products of when hydrogen peroxide is broken down by catalase?
So basically if you like it to be a statement instead of a question we can :)
Yes, I think I will choose a statement....can you make for me one? please
Well what do you expect that happens? What happens if you add hydrogen peroxide to catalase?
catalase breaks hydrogen peroxide down into oxygen and water; the reaction appears as white bubbles.
There you go. you just made your first hypothesis!!! :) "Catalase catalyze the reaction of hydrogen peroxide into molecular oxygen and water" Just a minor rewritten statement of what you said.
What you said there is exactly what a hypothesis should sound like. Well done.
Thank you thank you...just because with your help....so informative. So the hypothesis will be :" Catalase catalyze the reaction of hydrogen peroxide into molecular oxygen and water"
can we add if...then?
Yes. That is a excellent hypothesis. And I shall show you why in 2 sec. Well we can take that in two sec.
Yes..
Okay so with your hypothesis there are two things we gotta check: 1) hydrogen peroxide is made into water and oxygen. 2) catalase only catalyze the reaction. Now what does it mean that catalase catalyze the reaction?
Or let me ask like this: how does an enzyme work?
Hydrogen-PER-oxide breakdown: 2H2O2 --> 2H2O + O2
When a molecule of the right chemical for that enzyme comes along it will fit exactly into the shape. The area of particular shape is called the active site of the enzyme, as that is where the reaction takes place. The molecule that the enzyme works on is called the substrate.
Yeah not exactly what I meant, but close. [You are not copy pasting from the net are you ;)] No what I meant is that when an enzyme is speeding up a reaction it is NOT GETTING used during the reaction. This ability we call to catalyze.
oh..okay.
So what about the hypothesis start with "if...then"
Can you make one for me please? I can't think of anything at all. I'm stuck at here...:(
Enzyme Lab: When you complete the lab, make sure you are answering the questions about the liver. Don’t include the other foods you are testing yet. You will need to test one food, such as a potato, onion, or apple. This experiment goes on the “Enzyme Observation Form”. This form will contain your hypothesis, data, and conclusions about these additional foods.
So return to what matters: "2) catalase only catalyze the reaction." In order to test this, we expect that if we add hydrogen peroxide to the liver a reaction will happened. To show that catalase is a catalyst we will remove a part of the liver to a new glass and add more hydrogen peroxide, IF there is still activity, THEN we know catalase is a catalyst
"if we add hydrogen peroxide to the liver a reaction will happened. To show that catalase is a catalyst we will remove a part of the liver to a new glass and add more hydrogen peroxide, IF there is still activity, THEN we know catalase is a catalyst " So here is the hypothesis right? I think that's good :)
Good good :) (while you wrote I've been sitting writing the report my self. I am willing to share it with you, so long you can promise me it won't be turned it)
I think that the hypothesis is good as well! :)
OMG....I'm crying right now..Because I can say it in my entire life, there's no such good friend and helper like you. Thank you so much and yes I promise I won't turned it in. Please trust me...:) <3
So, you gonna share it using email right?
Ok...so we done with hypothesis part...right?
Yes we are done with the hypothesis I would say yes :)
And I am SUPER GLAD you made the hypothesis your self. Perfect! :D
Thanks...So we move to next questions. Deal?
Can you check for me the part "Purpose of the Lab" too?
Oh yes. I checked that and it is fine for me. But I think you should fast see if hydrogen peroxide is dangerous for the body since you say the liver break down toxic compounds. :)
What do you think? I don't know. Can you check it for me please? Plus, is this lab have to do something with antioxidant? I mean like I have research some of the lab and some they wrote down that: Purpose of this lab "It is trying to answer what the antioxidant does for the human body." Hypothesis In this lab we will test for the importance of antioxidant in the human body.My hypothesis for antioxidant is that it helps fight off free radicals.
Well... they are not exactly showing that. So I would consider it wrong to say.
And I have send a revised version to your email.
Ok....May I ask the report that you have sent it to me. There are some questions at the bottom that you don't fix it, so that's correct right?
Can we take that to the very end? And get all the other stuff done first? :)
Yes...sure
Analysis Analyze the data you collected in the lab Characterization of the reaction carried out in the liver: I suggest you do a table for your self O2 H2 C - - E + - Can you help me with this table. I don't know how to...make this data..
But try read what I wrote. And see if you like it, or you can use it.
It is so informative....I will have a closer look into it. Btw...many thanks. There's no more words so that I can say thank you to you.....:)
You are unsure what I meant with the analyze part?
Yes...I mean like the analyze the data part...
Well. The data analysis is based on 2 experiments: 1) the observation for hydrogen and/or oxygen. 2) the preserved ability to catalyze the reaction. 1) is the one with the table. And the table is based on the stick result. If it pops we know that hydrogen is +. if not we know it is - same goes for oxygen, if it glows more bright it is +, if not -
Can you do it for me please...i forgot it all.. sorry
Well I have in the document I send you?
Oh oh...I see... Sorry. Nevermind.. Ohh
Hehe no prob :)
Ok..No question. SO informative. Now, I think go to the Notebook Activity Ok? Are u ready??? My awesome, talented expert....<3
The notebook activity on the last page yes?
Yes, exactly...:)
