The theme is an important message that is central to your book. You should be able to find many things throughout your book that support the theme you choose. Conflict, events, characters, and their learning or changing—all these things can help you figure out the theme or themes of your book. When you think you have a good idea of what one main theme is, write a list of specific things (conflict, events, characters) that support your theme.
