@javk, some people aka myself don't distinguish vesicle and vacuole

yeah but you need to keep in mind that they are used for two different things, a vacuole will store water and nutrients, mostly sugars, whereas a vesicle transports proteins from (1) endoplasmic reticulum to the Golgi apparatus and (2) from the Golgi apparatus to the cell membrane. An easy way to remember this is vesicle sounds like vehicle which is also a transport medium :)
