If you mean during a science experiment; it's usually so that all the enzymes in the cell die thereby terminating both photosynthesis and respiration. This then acts as a control. In other words, you are killing the leaf so it doesn't do any biological activity during your experiment, so that your oxygen or carbon dioxide levels don't change. It also proves that nothing else in the leaf is causing any change.
