When a glass of cold water warms up to room temperature, that means that the molecules are converting the potential energy in their bonds to kinetic energy. Temperature is the average kinetic energy of molecules within a substance. Therefore, an increase in temperature means that molecules are beginning to move faster (gain KE), while an a decrease in temperature means that molecules are beginning to slow down (lose KE)
