Which is the strongest indication that your friend is going through a depression?

This is a bit of a vague question, but the major signs of depression are abnormal changes of mood, where you notice that someone is a bit irritable or angry, withdrawal from activities that they'd usually find fun, changes in how much they eat, restlessness/anxiety, and lack of energy. There's a lot more signs of depression but I'm not exactly sure what specifically you're looking for. Especially because depression does effect people differently.
