Sickle cell anemia is caused by a base substitution mutation. It's a change in the 6th codon for the beta chain of hemoglobin, which is changed from GAG to GTG on the non-coding DNA strand and on the mRNA codon it's changed from GAG to GUG. This results in a change in the amino acid produced since now Valine is produced instead of Glutamic acid.
