How do mutations within a cell occur?

I plan on doing a 2-3 minute presentations on how cancer develops in the body. I would like a few good links if you find any on how and why mutations occur.

I did something similar in my class last year but we did it on HIV in one of the units Aside from environmental factors like radiation and other carcinogens that impact cell integrity during gene expression some of the coding sequences are incorrect. Also during DNA replication if there are errors then it can cause mutations. Cells have defense systems to this such as the Nucleotide excision repair which removes incorrectly placed nucleotides with nuclease correction complexes. With regards to cancer, it is the absence of the property of DDI in cells. This could be due to the increase in protooncogenes or the lack of having tumor-suppressor genes (ie p53 which a major gene behind cancer).
