Well first of all proteins are composed of amino acids. Each amino acid is made up of an amino group and a carboxylic group, but each one has a differing "R" group. The bonds that hold or are used to hold amino acids together are called peptide bonds.
look at a simple example of key and lock, if key do not fits the lock it remains locked...i.e it remains nonfunctional. Come to next point--- there are millions of proteins and biological phenomenon occur due to interaction of molecules or proteins(here), so to have better proximity of molecule to another molecule which are in interaction should have a structure that fits. enzymes work on this principle broadly but there are examples where molecules change their structure when they come in proximity of another molecules! google it!:+)
