To find the y-intercept of an equation in the form of y=mx+b, would you substitute zero in for x?

Yes you want to find the co-ordinate where the line intercepts the y-axis. So x is always 0. The co-ordinate would be (0,y) so you plug that in and get y=m0 + b or y=b
