Does an enzyme use a substrate, and cause a reaction to make a different molecule, that is termed the product. Such as in the substrate glucose, in the first step in glycolysis?

Glycolysis comprises the 10 steps, which are catalysed by different enzymes. In the first step glucose is converted to glucose-6-phosphate by the enzyme:glucokinase or hexokinase. this enzyme is utilizing ATP and mg2+ ions.

To answer the first part of your question: yes. There can be a single substrate complex or a multiple ones. During their reaction with the enzyme on the active site, the subtrates within the complex creates a new chemical composition through the enzymatic mechamism (forming the product), but the enzyme itself doesn't change.
